What is the surest way to check to see that a URL has been indexed by Google?

Checking whether the site has been indexed in Google or not. Goto Google and type site:yourwebsitename.com.
this will show all the indexed pages of your website.

You can also use cache command to check when the crawlers came to your site.

My site is 2 years old and want some changes in its URL. Is it possible to redirect your URL? does it effects SEO.

You can go to Google and type site:"Your domain name" and enter. If your url is indexed, it will be appearing on the search results.
